Blood Partitioning

Certain drugs have the potential to permeate red blood cells leading to high blood to plasma ratios.The knowledge of such partitioning is important in understanding the compound’s pharmacokinetic behavior. Additionally assessment of the blood distribution of compounds allows for the determination the most appropriate matrix for analysis of the concentrations of compound in circulation.

Accumulation of compound in red blood cells may reduce the amount of compound available to cause the desired pharmacological effect.

Assay Methodology

At Quintara Discovery, blood distribution assays are routinely performed using fresh blood. The test compound is spiked in fresh whole blood and fresh plasma. After incubation at 37 °C for 1 hour, all samples are centrifuged, when plasma portion is produced from the spiked whole blood. Concentrations of the test compound are determined in both plasma produced from the spiked whole blood and the spiked plasma. The blood/plasma ratio is determined using the hematocrit value.
